讲座内容:

To date, non-invasive neuromodulation techniques have garnered significant attention due to their great potential in treating brain disorders and inspecting brain functions. In particular, transcranial ultrasound has emerged as a novel therapeutic modality owing to its high spatiotemporal resolution and non-invasive nature. However, the therapeutic efficacy of conventional transcranial ultrasound for treating diseases remains relatively limited. To address this, we developed a baicalein-loaded nanocarrier (BP-B) capable of responsive release of nitric oxide gas upon ultrasonic stimulation, thereby temporarily opening the blood–brain barrier and enhancing the efficiency of intracerebral drug delivery. Furthermore, the production of nitric oxide amplifies the ultrasound-induced activation of cortical neurons. In a mouse model of Parkinson’s disease, deep brain stimulation of the subthalamic nucleus (STN) with ultrasound combined with BP-B treatment significantly improved motor and cognitive functions and alleviated parkinsonian symptoms without inducing notable toxicity. Collectively, this combined pharmacological and ultrasound modulation strategy provides a new approach for the non-invasive treatment of neurodegenerative diseases.
